DETERMINATION OF TILIROSIDE IN PLANTS EXTRACTS
USING CHROMATOGRAPHICAL METHODS

Tiliroside is a widely-known acylated flavonol compound, first isolated from Tilia argentea Desf, whose structure was identified as kaempferol 3-O-b-D-(6\’\'-O-coumaroyl) glucopyranoside. Flavonoids are known to exhibit a broad spectrum of pharmacological activities, including antitumor, antimicrobial, antiviral,central vascular system and enzyme inhibiting activities. Tiliroside showed very potent anti-complement activity on the classical pathway of the complement system. The modulation of this activity plays an important part in inflammation. The compound was examined for its cytotoxic activity against a panel of human leucemic cell lines in vitro. It was active against two of the nine tested cell lines.
Tiliroside occurs commonly in the world of plants, e. g. in Rosaceae, Malvaceae, Tiliaceae, Platanaceae, Pteridaceae families. Because of positive pharmacological activities, the presence of this compound is desirable of great importance in medical plants.
In this work a reproducible, quick and sensitive method was elaborated for the separation and specific quantitation of tiliroside by isocratic RP-18-HPLC with ultraviolet detection.
Sample preparation was carried out by solid phase extraction with a recovery 96 %. After SPE separation all treatments of the eluents were quantitatively investigated on-line. The analysis of tiliroside was carried out using RP-18 column and 50 % methanol in 10 % trifluoroacetic acid as mobile phase. The combined SPE-HPLC method can be recommended for the screening analysis of the content of tiliroside in various plants organs as easy, rapid and reproducible chromatographic procedure.
